If you are tied up whilst sitting on a chair with one large length of rope by some laymen, is threr a very good chance that you can escape by making slack etc. Or is it actually possible to tie someone in this way securely?

If you have enough experience escaping from chairs you can work out how to tie somebody to a chair so they cannot get out. That is simply by remembering ideas that have made your escape harder and adapting them with the knowlege you have as an escapist. Another very experienced escapist might just be able to get out but probably knot. But no layman, even with tying experience, would be able to restrain a practised escaper. The knowledge comes from being restrained not doing the restraining.

It is all down to experience.

Depends on many factors.

How much rope?

How thick is the rope?

What kind of chair?

etc

Of course, it is likely you can escape in most cases; but is there going to be an audience there when you do is the better question.
